High Throughput Microfluidic Sample Preparation for Metagenomic Analysis

Metagenomics deal with analysis of genetic material from environmental samples. The ultimate goal is to reconstruct entire genomes of unknown microbial species. We present a sample preparation method using the Fluidigm© C1 microfluidic platform that integrates throughput of shotgun sequencing with bioinformatics simplicity of single cell microfluidics. Our method is tested with a benchmark samp...

Towards Point-of-care Diagnostics: a Microfluidic Sample Preparation Chip for Concentration of Bacteria and Rna Extraction

We report on the development and proof-of-concept of a microfluidic chip. It provides a full range of pretreatment steps for the point-of-care detection of bacteria with a total process time of less than 15 minutes. The device integrates concentration, lysis and nucleic acid purification. Successful operation is demonstrated using E. coli as a model organism.

Continuous-flow microfluidic blood cell sorting for unprocessed whole blood using surface-micromachined microfiltration membranes.

White blood cells (WBCs) constitute about 0.1% of the blood cells, yet they play a critical role in innate and adaptive immune responses against pathogenic infections, allergic conditions, and malignancies and thus contain rich information about the immune status of the body. Rapid isolation of WBCs directly from whole blood is a prerequisite for any integrated immunoassay platform designed for...

Microfluidic blood sample preparation for rapid sepsis diagnostics

Sepsis, commonly referred to as blood poisoning, is a serious medical condition characterized by a whole-body inflammatory state caused by microbial infection. Rapid treatment is crucial, however, traditional culturebased diagnostics usually takes 2-5 days. The overall aim of the thesis is to develop microfluidic based sample preparation strategies, capable of isolating bacteria from whole bloo...
